First off, you need to equip an item to your character. Go to the equip screen and select the defensive portion. Go down to the flask icon and select the item that you wish to equip. You can equip up to 3 of a particular item. Then when roaming the halls of the castle you can hold the left trigger then press the right trigger to use an item.
